Using a single depletion laser avoids laser misalignment and bleaching issues during stimulated emission depletion (STED) microscopy but limits the available fluorophores for use. Imaging with five fluorochromes using one depletion laser is possible using a fluorescence-lifetime imaging microscopy (FLIM)-STED approach to separate dyes with similar emission spectra but different lifetimes.
